He appeared with his lifelong friend James Cagney in nine films including Angels with Dirty Faces (1938) and Cagney's last film Ragtime (1981). O'Brien may be best remembered as Detective Mulligan opposite George Raft, Tony Curtis, Jack Lemmon and Marilyn Monroe in Some Like It Hot (1959) and the title role of the football coach in Knute Rockne, All American (1940) where he gave the speech to "win one for the Gipper" referring to a recently deceased football player, George Gipp, portrayed by a young Ronald Reagan.
